          R&D Systems为白细胞介素-17(IL-17)家族的配体和受体增添了新的荧光共轭抗体。白介素-17 家族的配体属于促炎细胞因子,能诱发局部细胞因子的产生,并参与调节免疫功能,其中最显著的是过敏性反应。 白介素-17家族的6位成员,IL-17,IL-17B,IL-17C,IL-17D,IL-17E(也称为IL-25)及IL-17F是一组在结构上相关联的蛋白质,在氨基酸C端附近折叠成一保守的半胱氨酸结,而氨基酸N端则属于可变区。白介素-17家族成员所激活的3种受体分别为IL-17受体,IL-17B 受体,和IL–17C受体。两个被命名为IL-17 RD 及IL-17 RE的孤儿受体与IL-17受体呈现同源性,但与其相应的功能配体,至今还未被人所知。
          利用新的共轭抗体,经流式细胞仪所检测出的IL-17B(图1),IL-17C(图2)和IL-17E(图3)的细胞内染色如下所示。IL-17B在脊髓上有高表达, 与IL-17B 受体相结合,但不与IL-17受体相结合。

    IL-21 and TGF-ß are required for differentiation of human TH17 cells

    The recent discovery of CD4+ T cells characterized by secretion of interleukin (IL)-17 (TH17 cells) and the naturally occurring regulatory FOXP3+ CD4 T cell (nTreg) has had a major impact on our understanding of immune processes not readily explained by the TH1/TH2 paradigm. TH17 and nTreg cells have been implicated in the pathogenesis of human autoimmune diseases, including multiple sclerosis, rheumatoid arthritis, inflammatory bowel disease and psoriasis1, 2. Our recent data and the work of others demonstrated that transforming growth factor-ß (TGF-ß) and IL-6 are responsible for the differentiation of naive mouse T cells into TH17 cells, and it has been proposed that IL-23 may have a critical role in stabilization of the TH17 phenotype3, 4, 5. A second pathway has been discovered in which a combination of TGF-ß and IL-21 is capable of inducing differentiation of mouse TH17 cells in the absence of IL-6 (refs 6–8). However, TGF-ß and IL-6 are not capable of differentiating human TH17 cells2, 9 and it has been suggested that TGF-ß may in fact suppress the generation of human TH17 cells 10. Instead, it has been recently shown that the cytokines IL-1ß, IL-6 and IL-23 are capable of driving IL-17 secretion in short-term CD4+ T cell lines isolated from human peripheral blood11, although the factors required for differentiation of naive human CD4 to TH17 cells are still unknown. Here we confirm that whereas IL-1ß and IL-6 induce IL-17A secretion from human central memory CD4+ T cells, TGF-ß and IL-21 uniquely promote the differentiation of human naive CD4+ T cells into TH17 cells accompanied by expression of the transcription factor RORC2. These data will allow the investigation of this new population of TH17 cells in human inflammatory disease.
