中科院两位学者发表最新文章解析胰腺癌新机制

胰腺癌是一种恶性程度很高、预后极差且诊断和治疗都很困难的消化道恶性肿瘤,而大多数的胰腺癌为胰腺导管腺癌(pancreatic ductal adenocarcinoma, PDAC)。
胰腺癌是一种恶性程度很高、预后极差且诊断和治疗都很困难的消化道恶性肿瘤,而大多数的胰腺癌为胰腺导管腺癌(pancreatic ductal adenocarcinoma, PDAC)。世界卫生组织2014年全球癌症研究报告显示,胰腺癌现已成为全球第七大致死性癌症,五年的存活率不到5%,在美国该类癌症为第四大致死性癌症。胰腺癌多发于发达国家,该病在我国的发病率处于较低水平。近十几年来,随着国民生活水平的不断提高,肉、蛋、奶等动物性食品在饮食结构中的比重不断提高,加之人们生活节奏和压力的加快、加大,胰腺癌的发生率处于一个上升趋势并维持在相对较高水平。由于我国人口基数大,每年我国胰腺癌的发生和死亡数占全球比重较高。
朊蛋白(Prion Protein, PrP)作为机体广泛表达的一种带有糖基磷脂酰肌醇锚(GPI-anchor)的糖蛋白,在中枢神经系统中有着高丰度的表达。该蛋白的异常可以导致包括人类在内的多种动物的神经退行性疾病,这包括众所周知的发生于上世纪80年代、流行于90年代英国的牛海绵状脑病(Bovine Spongiform Encephalopathy),又被称为“疯牛病”。过去很长一段时间对该蛋白的研究主要是从病原学和传染病学的角度。近十几年来越来越多的研究发现,朊蛋白在多种癌症中出现较高水平的表达,更为重要的是朊蛋白的表达对于多数癌症的发生、发展、预后及多重耐药具有很大的相关性。
中国科学院武汉病毒研究所研究员李朝阳早前对一系列PDAC细胞系的研究发现,PDAC细胞系出现PrP的高水平表达,且表达的PrP以前体缺陷的pro-PrP形式存在,即羧基端的GPI-PSS区没有被GPI-anchor所置换,而该区能够与细胞的内源性FLNa相互作用,从而引起细胞骨架和信号通路的改变,最终促进癌细胞的体外增殖、侵袭和体内生长。进一步的临床研究和调查发现,PrP的表达是胰腺癌病人预后差的生物学标志。
近期,生物化学研究杂志The Journal of Biological Chemistry 在线发表了武汉病毒所杨利恒、高振兴等的文章GPI Anchor Modification Machinery Deficiency is Responsible for the Formation of Pro-PrP in BxPC-3 and Increases Cancer Cell Motility,该研究由李朝阳指导,以两种胰腺导管腺癌细胞系(AsPC-1和BxPC-3)为研究对象,试图揭示BxPC-3胰腺癌细胞中原朊蛋白(pro-PrP)形成的机理,并进一步探讨前体缺陷型pro-PrP对癌细胞体外迁移的影响。结果显示,BxPC-3细胞中参与GPI-anchor生物合成、修饰的多个基因的异常特别是PIGF和PGAP1基因的低水平表达是导致pro-PrP形成的重要因素。更为重要的是,前体缺陷型的pro-PrP显然更有利于癌细胞的体外迁移。该研究首次证明了PrP的不同形式对癌细胞生理活动的影响,从而为包括胰腺癌在内的多种癌症的临床诊断、治疗和药物开发提供一定的理论基础。

GPI Anchor Modification Machinery Deficiency is Responsible for the Formation of Pro-PrP in BxPC-3 and Increases Cancer Cell Motility
The normal cellular prion protein (PrP) is a GPI-anchored, cell surface glycoprotein. But, in pancreatic ductal adenocarcinoma cell (PDAC) lines, such as BxPC-3, PrP exists as a pro-PrP retaining its glycosylphosphatidylinositol (GPI) peptide signaling sequence (GPI-PSS). Here, we report the identification of another PDAC cell line, AsPC-1, which expresses a matured, GPI-anchored PrP. Comparison of the 24 genes involved in the GPI anchor modification pathway between AsPC-1 and BxPC-3 revealed 15 of the 24 genes, including PGAP1 and PIG-F were down regulated in the latter cells. We also identified 6 missense mutations in DPM2, PIG-C, PIG-N and PIG-P alongside 8 silent mutations. When BxPC-3 cells were fused with Chinese Hamster Ovary (CHO) cells, which lack endogenous PrP, pro-PrP was successfully converted into matured, GPI-anchored PrP. Expression of the individual gene, such as PGAP1, PIG-F, or PIG-C into BxPC-3 cells does not result in PI-PLC sensitivity of PrP. However, when PIG-F but not PIG-P is expressed in PGAP1 expressing BxPC-3 cells, PrP on the cells surface becomes PI-PLC sensitive. Thus, low expression of PIG-F and PGAP1 are the major factors contributing to the accumulation of pro-PrP. More importantly, BxPC-3 cells expressing GPI anchored PrP migrate much slower than BxPC-3 cells bearing pro-PrP. In addition, GPI-anchored PrP bearing AsPC-1 cells also migrate slower than pro-PrP bearing BxPC-3 cells, although both cells express filamin A (FLNa). "Knocking out" PRNP in BxPC-3 cell drastically reduces its migration. Collectively, these results show that multiple gene irregularity in BxPC-3 cells is responsible for the formation of pro-PrP, and binding of pro-PrP to FLNA contributes to enhanced tumor cell motility.
